Is it possible to create a wall that has 2x6 top and bottom plates with 2x4 staggered studs?

Just a thought, build a wall with the right top and bottom then add 2x4 studs at half the spacing that are offset 0" then another set that is offset 2".  in the framing plan they probably won't line up properly and you'll have to edit them.  But atleast the count will be correct.

Exactly ...

As Dennis says, you can't define the stagger, but it can be edited in the framing plan, and the count is correct.
